Leclercia adecarboxylata C12, The Newly Isolated Cellulose-degrading Bacteria from Indonesian Coffee Pulp

Culturable cellulose-degrading microorganisms were collected from Arabica coffee pulp in East Java, Indonesia. Fifty isolates obtained, and thirty-three showed hydrolyzing zone on Carboxy Methyl Cellulose agar plates after Congo-Red staining. The highest specific CMCase activity was observed by C12, identified as Leclercia adecarboxylata based 16S-rRNA gene sequence analysis. SDS-PAGE of C12 cellulase revealed two bands with a molecular mass 95.49 81.28 kDa, respectively. Activity gel analysis the cellulolytic ability clear formation. optimal achieved at 50°C pH 9, retained 47% its initial incubation for 90 minutes. purified enzyme remains stable 5 to 10, 77% maximum activity. stimulated presence K+, Ca2+, Mg2+, Fe3+, while SDS EDTA reduced current study shows that thermostable-alkalophilic produced is very promising industrial applications.

A bacterial strain, PS4040, capable of degrading polycyclic aromatic hydrocarbons for use as the sole carbon source was isolated from oily-sludge-contaminated soil. The 16S rRNA gene showed 98.8% homology to that of Leclercia adecarboxylata. Comparative molecular typing with the clinical strain of L. adecarboxylata revealed that there were few comigrating and few distinct amplimers among them.
